Inhibition of Human Epidermal Growth Factor Receptor-2 (HER-2) from Pomelo (Citrus maxima) Flavonoid Compounds: an In Silico Approach

نویسندگان

چکیده

Citrus maxima or pomelo is a plant that has potential as an anticancer because it contains flavonoids. One of the targets breast receptors HER-2 protein. This research aims to determine activity, toxicity compound, and prediction physicochemical properties flavonoids contained in through silico approach. Flavonoid compounds were screened using SwissADME with Lipinski's rule five, Torsion, TPSA, P-Gp Non-Substrate. Compounds passed screening carried out molecular docking receptor (PDB ID: 3PP0) Molegro Virtual Docker (MVD). The (GDP was declared valid had RMSD<2Å. results showed there 11 flavonoid lower rerank score than comparison compound Trastuzumab. Toxicity predicted Protox II online tool safe limits, namely classes 5 3. Based on this research, can be concluded acacetin, diosmetin, honyucitrin, isosinensetin, nobiletin, sinensetin, tangeretin candidates for cancer drugs based natural ingredients. Keywords: cancer, maxima, HER-2, silico.

Breast Carcinoma; Human Epidermal Growth Factor Receptor-2 (HER-2) and Grading Correlation

Introduction: Overexpression of Human Epidermal Growth factor Receptor-2 (HER-2) is one of the most important prognostic and predictive factors of breast cancer, observed in 25% - 30% of breast carcinoma patients leading to poor prognosis and feasible anti HER-2 antibody drugs. The objective of this study was to evaluate the HER-2 frequency in target population and its correlat...

Human Epidermal Growth Factor Receptor 2 (HER-2/neu)-Directed Therapy for Rare Metastatic Epithelial Tumors with HER-2 Amplification

CASE 1 A 67-year-old Asian female was diagnosed with locally advanced high-grade salivary duct carcinoma in June 2011. Molecular analysis revealed human epidermal growth factor receptor 2 (HER-2) amplification. She received adjuvant therapy with carboplatin/paclitaxel/ trastuzumab and maintenance of trastuzumab. Upon disease progression, trastuzumab could not be continued due to lack of financi...

Effect of simultaneous inhibition of epidermal growth factor receptor and cyclooxygenase-2 in HER-2/neu-positive breast cancer.

PURPOSE HER-2/erbB2/neu is overexpressed in 25% to 30% of all invasive breast cancers and is associated with an aggressive course and reduced survival. HER-2/erbB2/neu breast tumors are frequently associated with up-regulation of cyclooxygenase (COX)-2 and activation of the epidermal growth factor receptor (EGFR) pathway, which promote enhanced cell growth and resistance to apoptosis. This stud...

Human Epidermal Growth Factor Receptor Family (HER) in Gastric Cancer

The epidermal growth factor receptor family consists of four members with similar structures: HER1/erbB1, HER2/erbB2, HER3/erbB3 and HER4/erbB4. The genes encoded for Erb are proto-oncogenes, or precursor genes, which under normal conditions are present in all cells of an organism. Proto-oncogenes can be activated in several ways, most often it is through structural changes of the cell genome s...

Expression of receptor tyrosine kinases epidermal growth factor receptor and HER-2/neu in synovial sarcoma.

BACKGROUND Synovial sarcomas are high-grade soft tissue neoplasms often characterized by a biphasic spindle and epithelioid cell morphology. The majority of synovial sarcomas harbor a specific chromosomal translocation in which the proximal portion of the SYT gene at chromosome 18q11 is fused to the distal portion of one of several duplicated SSX genes (most notably SSX1 and SSX2) at chromosome...
